Comparison of Immune Responses between Inactivated and mRNA SARS-CoV-2 Vaccines Used for a Booster Dose in Mice

Abstract

Background: The emergence of SARS-CoV-2 variants of concern (VOCs) has challenged the effectiveness of current COVID-19 vaccines. Booster doses are necessary to extend vaccine efficacy and enhance neutralization titers against these variants.

Methods: This study investigates the immune effects of mRNA vaccines based on the WT (prototypic strain) and omicron (B1.1.529) strains as booster doses in mice. With two-dose inactivated vaccine priming, mRNA boosters were evaluated for their ability to elevate IgG titers and cell-mediated immunity.

Results: Booster doses with mRNA vaccines improved immune protection against corresponding variants, but cross-protection against distinct strains remained limited.

Conclusion: The study reveals effective vaccination strategies against Omicron and future SARS-CoV-2 variants.
